This is a double-blind, randomised, placebo-controlled, double-dummy, parallel-group, multicentre, double-dummy, non-inferiority study, with a 6-month period of in-hospital treatment (NIT). All subjects had GAD diagnosed by using a screening questionnaire, and the primary outcome was the change from baseline in Hamilton Anxiety Rating Scale (HAM-A) scores.
The primary end point was the change from baseline in the HAM-A score. The secondary outcomes included treatment-emergent adverse events, use of the benzodiazepines, and adverse drug reaction profile (ADRS).
Twenty-two subjects with GAD were included in the present study, with a mean age of 59.7 years (range, 25-86 years), and seven subjects had been diagnosed with GAD between the ages of 25 and 59 years (mean age, 48.5 years). All subjects had participated in the previous 6 months of NIT, and the median time to first treatment with quetiapine was 5.7 months (range, 2-13 months).
Fourteen subjects (18.3%) completed the NIT, and five subjects (9.7%) completed the NIT 3 months after completion. There were no significant differences in the baseline HAM-A scores between the NIT and the placebo-treated groups (t-test, p=0.9).
Five subjects (6.4%) experienced the benzodiazepines, and four (7.5%) had a history of the benzodiazepine use.
Twenty-four subjects (63%) were taking the non-opioid tricyclic antidepressants (TCAs), and 11 (42%) had a history of the TCAs during the previous 6 months of NIT. The most common TCAs were amitriptyline (32.1%), clomipramine (32.5%), paroxetine (27.8%), citalopram (16.2%), imipramine (8.8%), fluoxetine (7.1%), paroxetine (5.8%), sertraline (5.9%), and venlafaxine (4.3%). None of the subjects had used anti-anxiety medications before taking the TCAs.
There were no significant differences in the baseline HAM-A scores between the NIT and the placebo-treated groups (t-test, p=0.8).
Quetiapine is not a potential treatment for generalized anxiety disorder in adults and adolescents with this condition. The NIT may provide a more effective alternative for treatment of symptoms of anxiety and panic disorders in this patient population.
